【中文摘要】 應用流式細胞分選儀(Flow cell sorter,FCS)和透射電子顯微鏡(Transmissional elec-tron microscope,TEM)等技術觀察了巴西蘑菇(Agaricus blazei)多糖誘導K562腫瘤細胞凋亡的試驗研究。試驗發現,流式細胞儀可檢測到經巴西蘑菇多糖誘導K562腫瘤96h后產生明顯的亞二倍體峰;電鏡下觀察到K562腫瘤細胞核固縮,核內染色質濃縮、凝聚,緊靠在細胞核膜邊沿,形成新月狀或環狀結構等典型的凋亡細胞的形態變化及凋亡小體。結果表明,巴西蘑菇多糖具有誘導K562腫瘤細胞凋亡的作用。
【英文摘要】 The research on the apoptosis of K562 tumor cells induced by polysaccharide extracted from Agaricus blazei was carried out using advanced technology of flow cell sorter and transmissional electron microscope.The result showed that,visible apoptotic peak could be detected by flow cell sorter after being induced for 96h by polysaccharide extracted from Agaricus blazei.Furthermore,karyopyknosis,chromatin condensed and aggregated,hugging the edge of cell nucleus,showing crico-structure,crescent-structure and other typical morphologic changes of apoptotic cell as well as apoptotic body could be seen in K562 cells through electro-microscope.It was concluded that the apoptosis of K562 tumor cells could be induced by polysaccharide extracted from Agaricus blazei.
